SATA3 standard max out about 600mb read and write. This is why you see almost all good SATA3 drive say 550/500 or something of the like.

The NVMe (PCIe) version of samsung ssds can carry more than 3.5 GB/s reads and 2.5GB/s writes. That is about 6X the speed of SATA3.

Imo if your just gaming and everyday use you don't really need more then sata3 /s write / read speed. I mean the difference it makes is almost not noticable in most cases. I haven't made the jump onto m.2 or PCIe cards yet. Haven't found it necessary or cost effective on my end. That's just my two cents.

Makes sense as a slimmer form factor for laptops

Price to performance, NVMe drives win, but honestly though I don't need something to be THAT fast, doubt for day to day usage I'd notice much of a difference, just rather want a cheap internal scratch disk. Paying twice the price for 6x the performance does have its appeal too though. Get whatever makes the most financial sense for you.

Keep in mind that most times it really does not "free" up a port as most desktop MB share the ports with M.2

The M2 form factor came about for tablets and laptops.

Are you talking about this?. It will be on lightning deal starting Sep 18, 7:20am. However, I'm not sure what will be the lightning deal price for tomorrow. I can tell you that the last 5 lightning deal price is $26.94.
