Not sure how Sims loads the different expansions, if it loads them all at once, or if you can only play one at a time.
Try defragmenting both drives, if you haven't.

Slow Performance Sims 2? Hards To Blame?
So I had sims 2 and a couple of the expansions on my 80GB hard drive with the windows installation.
Everything was working excellent at that point.
I added a second 250GB hard drive and added more expansions on the extra hard drive.
Now my games performance has gone down, with load times taking a while.
Is it because the game and expansions are over 2 hard drives, or is it because I installed some expansions the drive without the windows install?

So why didn't you install the games in your primary drive?

You should install them all on the same drive.
I would choose the 250GB, as it should be faster than the 80GB.
Load times are always slow for Sims2. The more expansion packs you add, the longer it takes to load as it does load items from the add-ons and expansions into the game.

Ya my gf, has a whole bunch loaded on hers. Took a long time to load.
I upgraded her laptop to 4gb of ram to help too.
But ya, I would do a fresh install of them on the 250GB, since it would be a faster driver in general over the 80gb._______________

Are these SATA drives or IDE? If they're IDE, are they both on the same cable? If they are, that could be a reason for slowness.

Use acronis trueimage or something to move everything from your 80gb to the 250. Using the new drive as your primary makes way more sense since it will be faster.
